Description
English: Supplementary Movie 1 Spatial-temporal changes in NR emission wavelengths in a fixed SH-SY5Y cell membrane. Localization metadata generated by sPAINT include localization (x and y coordinates), time (frame number) and spectral (▯) information. Hydrophobicity maps were generated by Nadaraya-Watson kernel regression on the spectral information retrieved at every point localization (i.e. spatial smoothing of numeric values observed at a set of irregular locations) and observed for a given time interval. Since the spectral information at fixed (or close in space) x and y-coordinates change with time, this calculation was made using a time-moving box of 200 over 3000 frames - to render the time/space change in local hydrophobicity. The temporal information of the hydrophobicity map was thus defined by the size of the moving box shift (16 frames) and was about 0.8 s. The frequency histogram of fluorescence emission peak corresponds in time to the data in the rendered membrane. The scale bar is 1 μm.
